This greatly affects the progress and cost of the experiment. 1. Inoculum volume of fermenter
First of all, we need to control the volume of strains, the right amount of strains can not only help the reproduction of strains in the tank, fermentation products quickly generated, but also to reduce the growth of miscellaneous bacteria. However, too much or too little will have an impact.
l The amount of nutrients and oxygen in the fermenter is fixed, and when the amount of strains is too large, it will easily lead to insufficient dissolved oxygen in the fermenter, and there will also be a large amount of metabolites, so that product synthesis is hindered.
l When there are too few strains of bacteria, the culture in the fermentation broth is insufficient, the fermentation time is delayed, and the productivity of the fermenter is greatly reduced.
So, how exactly do you determine the inoculum volume? Actually, it is also the volume of inoculated liquid and the volume of culture solution afterwards. Just determine it according to the specified percentage.
 
2. Fermenter inoculation methods
The second is the method of inoculation. For large industrial production, inoculation is usually done by pressure difference of the fermenter. For small fermenters, two other methods of inoculation are used: flame inoculation and syringe inoculation.

Step2 Regulate tank pressure of fermenter

Step3 Prepare a sterile syringe (inhalation of the seed liquid at the ultra-clean table in advance) and inject the seed liquid safely into the fermenter by aseptic operation.

This inoculation method can avoid contact between the strain and outside air, and avoid the use of open flame. It is suitable for small fermenters under 5L.
