Differences between triangle flasks and fermenters

In the fermentation process, there are usually two types of fermentation: triangular flask fermentation and fermenter fermentation. Triangular flask fermentation is experimented in the pre-fermentation stage. So what exactly is the difference between two ways?
 
1. The difference between triangular flasks and fermenters
  •  Inoculation
Triangular flask is the way to suck the strain into the flask with a test tube, and fermenters are usually inoculated aseptically by flame inoculation, etc.
  • Stirring
Triangle flask will be put into the machine to shake and turn the flask, which is less harmful to the shearing force of the strain.
Fermentation tank is by mechanical stirring, the shear force for the strain is larger, and the damage of the bacterium is much greater than the triangle flask.
  • DO
The surface of the triangular vial strain is in direct contact with the air, so DO is affected by the surrounding environment and dissolved oxygen cannot be controlled.
The fermentation tank can control the air passed in, and through stirring and aeration, the liquid and air are mixed evenly, and the dissolved oxygen parameters can be controlled.
  • pH
Triangle flask is controlled indirectly by buffer or calcium carbonate to control pH in the flask, which cannot be controlled precisely.
The pH of fermentation tank is adjusted by adding acid and alkali through direct flow, which can control its value within the set range.
  • Sterilization
Triangles are sterilized by outflow steam, while fermenters are sterilized by direct contact with steam .(see previous articles for specific sterilization methods)
  • Sampling
Triangle flask due to the small amount, inconvenient to take samples to test; fermentation tank can be sampled at any time to check the fermentation process.

2. Differences in fermentation results between triangular flasks and fermenters
Triangles can obtain experimental fermentation data more quickly under conditions such as limited space. However, fermenters are more accurate in the fermentation experimental data obtained because of their good confinement and controllable fermentation parameters. Triangular flask fermentation is also mainly to provide reference data for subsequent in-tank fermentation. Due to the different strain types, the fermentation method needs to be determined on a case-by-case basis.
